INSTRUMENTATION

Thermo-Finnigan LTQ Orbitraps (2)
The LTQ-Orbitrap mass spectrometer is a hybrid mass spectrometer comprised of a linear ion trap coupled to an orbitrap mass spectrometer. The orbitrap is a novel form of MS instrumentation with 60-100k resolution and <2ppm mass accuracy. The speed, dynamic range and sensitivity of this instrument make it ideal for proteomics experiments. The MPRC installed two LTQ-Orbitraps in February 2006 to increase the confidence and throughput of the facility.

Thermo-Finnigan LTQ
An automated electrospray ionization quadrupole linear ion trap instrument, characterized by high sensitivity and rapid, efficient MS/MS experiments, including multi-stage MS/MS (MSn). Its primary application is nano-scale liquid chromatography tandem mass spectrometry (nLC-MS/MS) for peptide identification from trypsin digests of proteins separated by 1D- and 2D- electrophoresis.

Thermo-Finnigan LTQ-FT
An automated hybrid instrument combining an LTQ analyzer with a 7 Tesla Fourier transform ion cyclotron resonance (FTICR) mass spectrometer. The FTICR analyzer provides mass resolving power up to 100,000 and mass accuracy of 1 part-per-million (ppm) or better for precursor ions, while the LTQ analyzer conducts MS/MS or MSn experiments in parallel. Its primary application is nLC-MS/MS for the identification of peptides, and characterization of post-translational modifications from either tryptic peptides or intact proteins.

Agilent LC/MSD TOF
An Agilent 1100 capillary liquid chromatograph interfaced to an electrospray ionization (ESI) time-of-flight (TOF) mass spectrometer. Mass resolving power of 10000, and mass accuracy better than 5 (ppm) for internally calibrated spectra. Its primary application is profiling and differentiating complex mixtures.

Waters Q-TOF-API US
An automated quadrupole-time-of-flight mass spectrometer, for automated nano-scale liquid chromatography tandem mass spectrometry (nLC-MS/MS). Mass resolving power of 10,000 for precursor and product ions with mass accuracies of 5-10 ppm using a lock mass. Data-dependent selection of precursor ions. Its primary application is nLC-MS/MS for peptide identification from either in-gel digestions or shotgun digestions.

Waters Q-TOF-2
A quadrupole-time-of-flight mass spectrometer, for automated nano-scale liquid chromatography tandem mass spectrometry (nLC-MS/MS). It has mass resolving power of 8,000 for precursor and product ions and data-dependent selection of precursor ions. Its primary application is nLC-MS/MS for peptide identification from either in-gel digestions or shotgun digestions.

Thermo-Finnigan TSQ Quantum Ultra
A triple stage quadrupole mass spectrometer interfaced to liquid chromatography via an electrospray ionization interface (ESI). Its primary application is quantification of targeted analytes using tandem mass spectrometry (MS/MS) and internal standards labeled with stable isotopes.

Waters LCT Mass Spectrometer
An electrospray ionization time-of-flight mass spectrometer with mass resolving power of 5000. Its primary application is general molecular weight determinations of proteins and peptides and characterizing non-covalent interactions.

Bio-Rad Biologic FPLC System (2)
The BioRad DuoFlow and Pathfinder FPLC (fast performance liquid chromatography) systems allow for purification of samples using over 30 different chromatography matrices. These techniques include, but are not limited to: ion exchange, size exclusion, affinity, antibody purification and desalting. The systems can be used with a refrigerated chromatography cabinet and contain analytical software to integrate and overlay chromatograms for sample comparison.

APEX 396 Multiple Peptide Synthesizer (2)
A synthetic peptide synthesizer used for the assembly of synthetic peptides by orthogonal Fmoc solid phase peptide chemistry (SPPS). The instrument has the capability of synthesizing up to 48 peptide sequences simultaneously at small scale (40 μmol). The MPRC has two APEX 396 instruments in operation.

ABI 433A Peptide Synthesizer (3)
A synthetic peptide synthesizer used for the assembly of synthetic peptides by orthogonal Fmoc solid phase peptide chemistry (SPPS). The instrument is used for the synthesis of long peptides (i.e.,> 50 amino acids) at 100, 250 or 500 umol scales. Currently the MPRC has three ABI 433A instruments in operation.

Thermo-Finnigan MSQ
A single quadrupole mass spectrometer that contains an electrospray ionization interface for the introduction of samples collected from HPLC fractions. It has a mass resolving power of 5000 and mass accuracy of about 50 ppm. Its primary application is for mass determination of synthetic peptides for quality control of peptide synthesis services.

ABI Procise 492 Protein Sequencer
An automated protein sequencer for the de novo sequencing of peptides by Edman N-terminal chemistry that are usually isolated by capillary LC (ABI 173 MicroBlotter ) in the MPRC. Resultant PTH amino acids are analyzed and identified by on-line chromatography utilizing ABI software for peptide sequence determination. The MPRC has two Procise instruments, which operate at two levels of sensitivity (10-50 pmol and < 1 pmol) for sample analysis.

BioRad PROTEAN IEF Cells
The MPRC has three PROTEAN Cells for the separation of complex protein mixtures by isoelectric focusing (IEF). This first step is required for the separation of proteins by 2D-PAGE. Each unit utilizes immobilized pH gradient (IPG) strips (choice of two lengths; varying pH gradients) for the separation of samples by IEF, and can accommodate up to 12 different samples in a single IEF run.

BioRad PROTEAN Plus & Dodeca Chambers
The MPRC has three PROTEAN Dodeca chambers of varying size for the separation of protein mixtures in the second dimension by SDS polyacrylamide gel electrophoresis (PAGE). Each dodeca chamber allows for the separation of 4 samples (in triplicate) in a single 2D PAGE run. The MPRC offers its 2D-PAGE service utilizing cast or pre-cast gels in three different sizes (BioRad mini, Criterion, and Large Format 20 x 20 cm).
